Rapid, easy-to-use, filter-based technology for the enrichment of mononuclear cells
Pall’s novel Purecell Select System revolutionizes the enrichment of mononuclear cells (MNC) with a new filtration-based platform that is fast, easy to use, and produces consistently high cell recoveries. The system frees scientists from traditional lab-based methodologies such as density gradient centrifugation that are known to be time consuming and labor intensive.- Accelerated processing – Achieve enrichment of MNC from 20 to 120 mL of whole blood in 15 minutes or less.
- Easy to use – Simply transfer the sample to the sample input bag, filter the sample, then back-flush the Harvest Solution through the filter, collecting the enriched MNC in a collection bag. During the filtration process, the majority of the plasma, red blood cells, and platelets pass through the filter into the waste bag below. The MNC are captured on the filter and released during the back-flush process with the optimized Harvest Solution. No capital equipment or special training is required.
- Consistent results – With the Purecell Select System, the uniquely designed filter and optimized Harvest Solution do all the difficult cell separation work. This results in a highly robust process with consistent results, critical when working with highly variable biologic samples. Using a filtration-based platform minimizes operator training and variability between operators, resulting in a more predictable cell yield.
- Higher cell recovery – MNC recovery from 50 mL whole blood consistently averages > 70%. The Purecell Select System consistently recovers 20 to 50% more MNC than Ficoll* processing.
- Increased level of safety – The risk of sample contamination and biohazard exposure is reduced in this smart-set design by incorporating needleless access ports and eliminating open-tube processing.

A Quick Look at This Fast, High-Yield Alternative to Traditional Density Gradient Centrifugation
Pall’s Purecell Select System is an alternative technology to density gradient centrifugation for the enrichment of MNC from whole blood or cord blood. Many density separation media are available commercially, of which Ficoll-Paque* is the most widely used.
Pall’s filtration-based system provides researchers with an alternative enrichment technology that is significantly faster than traditional methods and results in a higher cell yield than Ficoll separations. The MNC recovery from the Purecell Select System typically averages > 70% (Figure 1). MNC recovery across the WB volume range processed by the Pall system (20 to 120 mL) typically averages > 65% (data not shown).
Percent Recovery of Major Leukocyte Populations

Percent recovery of major cell types in 50 mL WB starting material, Purecell Select System processed samples and Ficoll samples. Percent recovery is calculated based on WBC, platelet and red blood cell (RBC) concentration (Cell-Dyn* 1800); percent lymphocytes, monocytes and granulocytes is based on phenotype data (BD FACSCalibur system). Duplicates are averaged. Means +/- 1 SD from 6 donors are reported.
Phenotypic analysis of processed samples shows that the Purecell Select System yields more cells of all major types than are recovered with Ficoll separations (Figure 2). The cell population from the Purecell Select System differs from the cell population recovered from Ficoll in that the cell population from the Pall system has a higher concentration of granulocytes and red blood cells. Researchers will need to assess the specific requirements of their applications. Protocol modifications can reduce the residual red blood cell concentration.Total Cell Number of Major Leukocyte Populations
